Sure you need license. We have 4 licenses according to engine displacement volume of motorbike.
1, 50cc or less
2, 125cc or less
3, 400cc or less
4, no limit
Only 1 does not have driving test (written exam only). You have to take both written and driving tests for 2,3 and 4.
I don’t think it’s realistic for you to take license unless you can read Japanese. The written test is in Japanese.

To get a motorbike licence, there are different licences for different bikes. I took the no limit one and it cost me about 180,000 Yen. It took about 10 weeks to get it. To ride a scooter, a normal car licence is enough. If you come here as a tourist, you can use your American licence with an International licence. If you are staying longer than 12 months, you need to change your American licence to a Japanese one. Unfortunately, America doesn’t have an agreement with Japan, so you will need to take some written and practical tests which you can take in English.
